Macrocycle Synthesis [ back to DPC Platform/Technology ]

Ensemble is using DPC™ to make large numbers of macrocyclic Ensemblins. These are attractive molecules for binding to drug discovery targets with extended binding motifs, as the macrocycles present a number of binding groups from a pre-organized cyclic framework.

DPC is especially good for making macrocycles, as the final cyclization occurs with a high degree of control. In conventional synthesis competition between the intramolecular reaction, which gives rise to the desired macrocyclic product, and intermolecular reactions produces unwanted reaction products. By using DPC, the DNA isolates the precursors from intermolecular reactions, resulting in efficient conversion to the macrocycle products.

Example of an Ensemblin™

We have a number of different backbone structures available, and combined with variation of the building blocks used in compound synthesis, we have access to billions of potential Ensemblin structures.
